ABSTRACT The Caatinga biome features an exclusive endemic biodiversity, and is characterized by the presence of xerophytic, deciduous vegetation, high temperatures, and low rainfall. This important park has undergone anthropization, especially through extraction of firewood and timber and growing plants for raising goats. The objectives of this study were to compare the communities of filamentous fungi present in the preserved area and in the anthropized soil of the Catimbau National Park in Buíque, PE, Brazil, and to evaluate the impacts of anthropization on such communities. In this study we evaluated the ability of Saccharomycopsis schoenii Nadson and Krassiln (UWO-PS 80-91) as biocontrol agent against plant pathogenic filamentous fungi P. expansum Link (UFMG 01-2002), P. italicum Wehmer (LCP 61.1199), and P. digitatum (Pers.: Fr.) (LCP 984263, LCP 68175 and LCP 4354). S. schoenii was able to reduce disease severity in oranges inoculated with all fungi. Among the phytopathogens, P. digitatum LCP4354 was the most virulent whereas P. digitatum LCP 68175 was the most susceptible to predation. The yeast was able to survive for 21 days on the fruit surface and did not produce lesions on oranges. Production of antagonistic substances by S. schoenii was not detected using standard techniques. Cette étude s’est inscrite dans un contexte d’évaluation de la salubrité des mollusques bivalves destinés à la consommation humaine. Ces organismes peuvent en effet accumuler des toxines, en particulier de microalgues, en concentrations importantes, pouvant induire des intoxications chez les personnes les consommant. Afin d’assurer la protection des consommateurs, les toxines réglementées sont recherchées dans les coquillages par techniques physico-chimiques. In order to assess the putative toxigenic risk associated with the presence of fungal strains in shellfish-farming areas, Penicillium strains were isolated from bivalve molluscs and from the surrounding environment, and the influence of the sample origin on the cytotoxicity of the extracts was evaluated. Extracts obtained from shellfish-derived Penicillia exhibited higher cytotoxicity than the others. En la actualidad, es altamente necesario encontrar nuevos y más seguros productos agroquímicos. En este sentido, los micromicetos son una fuente importante de productos naturales los cuales pueden ser usados en el control de enfermedades vegetales. Con el objetivo de contribuir a la búsqueda de productos con aplicaciones antimicrobianas, un total de 49 cepas fúngicas fueron aisladas de dos cenotes de la península de Yucatán. Our investigation of integrated biological control (IBC) started with an assay testing activity of the predacious yeast Saccharomycopsis crataegensis UFMG-DC19.2 against Penicillium digitatum LCP 4354, a very aggressive fungus that causes postharvest decay in oranges. Under unfavourable environmental conditions, the yeast showed a high potential for control (39.9% disease severity reduction) of this fungus. This result was decisive for the next step, in which S. crataegensis was tested in association with sodium bicarbonate salt, a generally regarded as safe (GRAS) substance. Protoplast fusion between complementary auxotrophic and morphological mutant strains of Penicillium griseoroseum and P. expansum was induced by polyethylene glycol and calcium ions (Ca2+). Fusant strains were obtained in minimal medium and a prototrophic strain, possibly diploid, was chosen for haplodization with the fungicide benomyl. Different recombinant strains were isolated and characterized for occurrence of auxotrophic mutations and pectinolytic enzyme production. The fungus Penicillium purpurogenum was found to produce intracellular ß-glucosidase. Maximum activity of ß-glucosidase was observed on sucrose. Various cultural parameters of cultivation of P. purpurogenum for production of ß-glucosidase were optimized. Maximum enzyme content was observed after 96 hours of cultivation at 30º;C. Addition of amino acids histidine and cysteine induced ß-glucosidase synthesis to certain extent. The optimum temperature and pH for ß-glucosidase activity was 50º;C and 5.5 respectively. ß-glucosidase of P.purpurogenum shows stability at pH 2 thus it could be an ideal enzyme for debittering in fruit juice and wine industries. |
